This sequence change replaces alanine with threonine at codon 2615 of the SETX protein (p.Ala2615Thr). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and threonine. This variant has been observed in individual(s) with clinical features of amyotrophic lateral sclerosis (Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SETX protein function.
