This variant lies in the LRP4 gene (transcript NM_002334.4) at coding-DNA position 4148, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 1383 with glycine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with glycine at codon 1383 of the LRP4 protein (p.Glu1383Gly). The glutamic acid residue is highly conserved and there is a moderate physicochemical difference between glutamic acid and glycine. This variant is present in population databases (rs202151304, ExAC 0.006%). This variant has not been reported in the literature in individuals with LRP4-related conditions.
