Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001330360.2(POLA1):c.3607C>G (p.Gln1203Glu), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt POLA1 protein function. ClinVar contains an entry for this variant (Variation ID: 1366382). This variant has not been reported in the literature in individuals affected with POLA1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 1197 of the POLA1 protein (p.Gln1197Glu). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
