Uncertain significance for Epileptic encephalopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004104.5(FASN):c.2452C>T (p.Pro818Ser), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs762145412, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This sequence change replaces proline, which is neutral and non-polar, with serine, which is neutral and polar, at codon 818 of the FASN protein (p.Pro818Ser). This variant has not been reported in the literature in individuals affected with FASN-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. ClinVar contains an entry for this variant (Variation ID: 1365783).
