This variant lies in the MYBPC3 gene (transcript NM_000256.3) at the canonical splice donor site of the intron immediately after coding-DNA position 3814, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.3814+1G>C intronic variant results from a G to C substitution one nucleotide after coding exon 33 of the MYBPC3 gene. This variant was reported in individual(s) with features consistent with hypertrophic cardiomyopathy (Hag&egrave;ge A et al. Int J Cardiol. 2025 Jan;419:132692; Ambry internal data). Variants that disrupt the canonical splice site are expected to result in aberrant splicing. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. A resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNA decay; although, direct evidence is unavailable.
