Uncertain significance for Primary ciliary dyskinesia 32 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_031924.8(RSPH3):c.-362A>G, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The cysteine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with RSPH3-related conditions. This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 22 of the RSPH3 protein (p.Tyr22Cys). This variant is not present in population databases (gnomAD no frequency).
