This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 1529, where T is replaced by C; at the protein level this means replaces leucine at residue 510 with proline — a missense variant. Submitter rationale: This sequence change replaces leucine with proline at codon 510 of the DSP protein (p.Leu510Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with DSP-related conditions.
