Uncertain significance for 3-methylglutaconic aciduria with deafness, encephalopathy, and Leigh-like syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032861.4(SERAC1):c.772C>T (p.Pro258Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with SERAC1-related conditions. This variant is present in population databases (rs571113650, ExAC 0.009%). This sequence change replaces proline with serine at codon 258 of the SERAC1 protein (p.Pro258Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine.
