Uncertain significance for Hypomyelinating leukodystrophy 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006087.4(TUBB4A):c.475_476inv (p.Phe159Asn), citing Invitae Variant Classification Sherloc (09022015): The frequency data for this variant in the population databases is not available, as this variant may be reported as separate entries in the ExAC database. This sequence change replaces phenylalanine with asparagine at codon 159 of the TUBB4A protein (p.Phe159Asn). The phenylalanine residue is moderately conserved and there is a large physicochemical difference between phenylalanine and asparagine. This variant has not been reported in the literature in individuals affected with TUBB4A-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). This missense change has been observed in at least one individual who was not affected with TUBB4A-related conditions (Invitae).
