Likely pathogenic for Distal myopathy with posterior leg and anterior hand involvement; Myofibrillar myopathy 5; Hypertrophic cardiomyopathy 26 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001458.5(FLNC):c.5683G>C (p.Ala1895Pro), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the FLNC gene (transcript NM_001458.5) at coding-DNA position 5683, where G is replaced by C; at the protein level this means replaces alanine at residue 1895 with proline — a missense variant. Submitter rationale: In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on FLNC protein function. This sequence change replaces alanine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 1895 of the FLNC protein (p.Ala1895Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with restrictive cardiomyopathy (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1364549).
