Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.623A>G (p.Tyr208Cys), citing Ambry Variant Classification Scheme 2023: The p.Y208C variant (also known as c.623A>G), located in coding exon 5 of the RECQL gene, results from an A to G substitution at nucleotide position 623. The tyrosine at codon 208 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
