Uncertain significance for Developmental and epileptic encephalopathy, 25 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_177550.5(SLC13A5):c.1094C>T (p.Thr365Ile), citing Invitae Variant Classification Sherloc (09022015): The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This sequence change replaces threonine with isoleucine at codon 365 of the SLC13A5 protein (p.Thr365Ile). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant has not been reported in the literature in individuals affected with SLC13A5-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
