Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030962.4(SBF2):c.3565A>G (p.Arg1189Gly), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with SBF2-related conditions. This variant is present in population databases (rs769777008, ExAC 0.01%). This sequence change replaces arginine with glycine at codon 1189 of the SBF2 protein (p.Arg1189Gly). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and glycine.
