Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 1363766). This variant has not been reported in the literature in individuals affected with TMTC3-related conditions. This variant is present in population databases (rs780546810, gnomAD 0.1%), including at least one homozygous and/or hemizygous individual. This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 239 of the TMTC3 protein (p.Leu239Pro).
