Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001197104.2(KMT2A):c.10350C>G (p.Asp3450Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with glutamic acid at codon 3450 of the KMT2A protein (p.Asp3450Glu). The aspartic acid residue is weakly conserved and there is a small physicochemical difference between aspartic acid and glutamic acid. This variant is present in population databases (rs781903452, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with KMT2A-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt KMT2A protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
