Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001197104.2(KMT2A):c.10405C>A (p.His3469Asn), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with KMT2A-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces histidine with asparagine at codon 3469 of the KMT2A protein (p.His3469Asn). The histidine residue is weakly conserved and there is a small physicochemical difference between histidine and asparagine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt KMT2A protein function.
