Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002979.5(SCP2):c.838A>G (p.Met280Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 280 of the SCP2 protein (p.Met280Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs751880143, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with SCP2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C15"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
