Uncertain significance for COG5-congenital disorder of glycosylation — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006348.5(COG5):c.2204G>T (p.Ser735Ile), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with COG5-related conditions. This sequence change replaces serine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 766 of the COG5 protein (p.Ser766Ile). This variant is present in population databases (no rsID available, gnomAD 0.0009%). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
