Uncertain significance for BENTA disease; Severe combined immunodeficiency due to CARD11 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032415.7(CARD11):c.3213dup (p.Ile1072fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ile1072Hisfs*9) in the CARD11 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 83 amino acid(s) of the CARD11 protein. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CARD11-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
