Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002095.6(GTF2E2):c.103T>A (p.Ser35Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with threonine, which is neutral and polar, at codon 35 of the GTF2E2 protein (p.Ser35Thr). This variant has not been reported in the literature in individuals affected with GTF2E2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1361940). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (gnomAD no frequency).
