This variant lies in the CASR gene (transcript NM_000388.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 186, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects an acceptor splice site in intron 2 of the CASR gene. RNA analysis indicates that this variant induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). Disruption of this splice site has been observed in individual(s) with clinical features of familial hypocalciuric hypercalcemia (PMID: 11668634). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 11668634). For these reasons, this variant has been classified as Pathogenic.
