NM_005431.2(XRCC2):c.341G>A (p.Arg114Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R114K variant (also known as c.341G>A), located in coding exon 3 of the XRCC2 gene, results from a G to A substitution at nucleotide position 341. The arginine at codon 114 is replaced by lysine, an amino acid with highly similar properties. This alteration has been reported in 0/13087 breast cancer cases and 1/5488 control individuals in the UK (Decker B et al. J Med Genet, 2017 11;54:732-741). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
