Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080473.5(GATA5):c.721C>T (p.Leu241Phe), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with GATA5-related conditions. This variant is present in population databases (rs781965965, ExAC 0.04%). This sequence change replaces leucine with phenylalanine at codon 241 of the GATA5 protein (p.Leu241Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
