Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001352754.2(ARMC9):c.622G>C (p.Glu208Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with glutamine at codon 208 of the ARMC9 protein (p.Glu208Gln). The glutamic acid residue is moderately conserved and there is a small physicochemical difference between glutamic acid and glutamine. This variant also falls at the last nucleotide of exon 6, which is part of the consensus splice site for this exon. This variant is present in population databases (rs753678888, ExAC 0.02%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Experimental studies and prediction algorithms are not available or were not evaluated, and the effect of this variant on mRNA splicing is currently unknown. This variant has not been reported in the literature in individuals affected with ARMC9-related conditions.
