Pathogenic for Cranioectodermal dysplasia 2; Short-rib thoracic dysplasia 7 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020779.4(WDR35):c.1954_1955insAAAC (p.Leu652fs), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Asp661Asnfs*6) in the WDR35 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WDR35 are known to be pathogenic (PMID: 22486404, 29068549). This variant has not been reported in the literature in individuals affected with WDR35-related conditions. For these reasons, this variant has been classified as Pathogenic.
