Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005502.4(ABCA1):c.975del (p.Trp324_Tyr325insTer), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ABCA1 gene (transcript NM_005502.4) at coding-DNA position 975, deleting one base. Submitter rationale: Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic. This variant has not been reported in the literature in individuals affected with ABCA1-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr325*) in the ABCA1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ABCA1 are known to be pathogenic (PMID: 10525055, 10760292, 20880529).
