This variant lies in the DMD gene (transcript NM_004006.3) at coding-DNA position 429, where G is replaced by T; at the protein level this means replaces tryptophan at residue 143 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tryptophan with cysteine at codon 143 of the DMD protein (p.Trp143Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine. This variant is present in population databases (rs763567086, ExAC 0.01%). This variant has not been reported in the literature in individuals with DMD-related conditions.
