This variant lies in the IL12RB1 gene (transcript NM_005535.3) at coding-DNA position 1750, where C is replaced by T; at the protein level this means replaces proline at residue 584 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been observed in individual(s) with clinical features of Mendelian susceptibility to mycobacterial disease (Invitae). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with serine at codon 584 of the IL12RB1 protein (p.Pro584Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine.
