Uncertain significance for Mitochondrial hypertrophic cardiomyopathy with lactic acidosis due to MTO1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012123.4(MTO1):c.85A>T (p.Ser29Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with cysteine at codon 29 of the MTO1 protein (p.Ser29Cys). The serine residue is weakly conserved and there is a moderate physicochemical difference between serine and cysteine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with MTO1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
