This variant lies in the PRIMA1 gene (transcript NM_178013.4) at coding-DNA position 163, where C is replaced by T; at the protein level this means replaces arginine at residue 55 with tryptophan — a missense variant. Submitter rationale: This sequence change replaces arginine with tryptophan at codon 55 of the PRIMA1 protein (p.Arg55Trp). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs146599561, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with PRIMA1-related conditions.
