NM_000377.3(WAS):c.149T>A (p.Val50Asp) was classified as Uncertain significance for Wiskott-Aldrich syndrome; X-linked severe congenital neutropenia; Thrombocytopenia 1 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces valine with aspartic acid at codon 50 of the WAS protein (p.Val50Asp). The valine residue is highly conserved and there is a large physicochemical difference between valine and aspartic acid. This variant has not been reported in the literature in individuals with WAS-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Not Available").
