This variant lies in the ABCA4 gene (transcript NM_000350.3) at coding-DNA position 2549, where A is replaced by G; at the protein level this means replaces tyrosine at residue 850 with cysteine — a missense variant. Submitter rationale: Variant summary: ABCA4 c.2549A>G (p.Tyr850Cys) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251412 control chromosomes (gnomAD). c.2549A>G has been reported in the literature in multiple individuals affected with Stargardt Disease (e.g. Oldani_2012, Khan_2020, Cornelis_2022), although in some cases it was reported as part of a complex allele with a likely pathogenic variant c.5882G>A (p.Gly1961Glu) in cis (Lee_2021). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
