NM_002439.5(MSH3):c.1173+1G>A was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1173+1G>A intronic variant results from a G to A substitution one nucleotide after coding exon 7 of the MSH3 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, in silico splice site analysis for this alteration is inconclusive. This nucleotide position is highly conserved in available vertebrate species. Based on the available evidence, the clinical significance of this alteration remains unclear.
