Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.7729T>C (p.Tyr2577His), citing Ambry Variant Classification Scheme 2023: The p.Y2556H variant (also known as c.7666T>C), located in coding exon 51 of the NF1 gene, results from a T to C substitution at nucleotide position 7666. The tyrosine at codon 2556 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
