Uncertain significance for Nemaline myopathy 10 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_198271.5(LMOD3):c.76A>G (p.Asn26Asp), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with LMOD3-related conditions. This variant is present in population databases (rs541421744, ExAC 0.01%). This sequence change replaces asparagine with aspartic acid at codon 26 of the LMOD3 protein (p.Asn26Asp). The asparagine residue is moderately conserved and there is a small physicochemical difference between asparagine and aspartic acid. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
