Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172362.3(KCNH1):c.630C>G (p.Ile210Met), citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt KCNH1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with KCNH1-related conditions. This sequence change replaces isoleucine with methionine at codon 210 of the KCNH1 protein (p.Ile210Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is not present in population databases (ExAC no frequency).
