Uncertain significance for Myopathy, tubular aggregate, 2; Combined immunodeficiency due to ORAI1 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000012.12:g.121641235G>T, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with aspartic acid at codon 166 of the ORAI1 protein (p.Glu166Asp). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ORAI1-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
