Uncertain significance for Cranium bifidum occultum — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002449.5(MSX2):c.77C>T (p.Pro26Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 26 of the MSX2 protein (p.Pro26Leu). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This variant has not been reported in the literature in individuals affected with MSX2-related conditions. This missense change has been observed in at least one individual who was not affected with MSX2-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 1357486). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
