Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.94G>A (p.Glu32Lys), citing Ambry Variant Classification Scheme 2023: The p.E32K variant (also known as c.94G>A), located in coding exon 1 of the AXIN2 gene, results from a G to A substitution at nucleotide position 94. The glutamic acid at codon 32 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
