Uncertain significance for Autosomal recessive distal spinal muscular atrophy 1; Charcot-Marie-Tooth disease axonal type 2S — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002180.3(IGHMBP2):c.1564A>G (p.Ile522Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the IGHMBP2 gene (transcript NM_002180.3) at coding-DNA position 1564, where A is replaced by G; at the protein level this means replaces isoleucine at residue 522 with valine — a missense variant. Submitter rationale: Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt IGHMBP2 protein function. This variant has not been reported in the literature in individuals with IGHMBP2-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with valine at codon 522 of the IGHMBP2 protein (p.Ile522Val). The isoleucine residue is moderately conserved and there is a small physicochemical difference between isoleucine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
