Uncertain significance for Atypical hemolytic-uremic syndrome — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_172351.3(CD46):c.191G>A (p.Cys64Tyr), citing Genomenon Sequence Variant Interpretation Standards. This variant lies in the CD46 gene (transcript NM_172351.3) at coding-DNA position 191, where G is replaced by A; at the protein level this means replaces cysteine at residue 64 with tyrosine — a missense variant. Submitter rationale: CD46 p.Cys64Tyr (c.191G>A) is a missense variant that changes the amino acid at residue 64 from Cysteine to Tyrosine. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:29511899;35619721). It is absent or not present at a significant frequency in gnomAD. In silico models agree that this variant is possibly or probably damaging. In conclusion, we classify CD46 p.Cys64Tyr (c.191G>A) as a variant of uncertain significance.
