Uncertain significance for Autosomal recessive DOPA responsive dystonia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000360.4(TH):c.1459G>C (p.Asp487His), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TH protein function. This variant has not been reported in the literature in individuals with TH-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with histidine at codon 518 of the TH protein (p.Asp518His). The aspartic acid residue is weakly conserved and there is a moderate physicochemical difference between aspartic acid and histidine.
