Uncertain significance for Deficiency of malonyl-CoA decarboxylase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012213.3(MLYCD):c.208G>A (p.Asp70Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with asparagine at codon 70 of the MLYCD protein (p.Asp70Asn). The aspartic acid residue is weakly conserved and there is a small physicochemical difference between aspartic acid and asparagine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with MLYCD-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
