Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001292063.2(OTOG):c.6928C>A (p.Arg2310Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with serine at codon 2322 of the OTOG protein (p.Arg2322Ser). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and serine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This variant has not been reported in the literature in individuals with OTOG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
