NM_005228.5(EGFR):c.89T>A (p.Val30Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V30D variant (also known as c.89T>A) is located in coding exon 2 of the EGFR gene. The valine at codon 30 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This change occurs in the first base pair of coding exon 2. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
