Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_000059.4(BRCA2):c.6278A>G (p.His2093Arg), citing Quest Diagnostics criteria. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 6278, where A is replaced by G; at the protein level this means replaces histidine at residue 2093 with arginine — a missense variant. Submitter rationale: The BRCA2 c.6278A>G (p.His2093Arg) variant has been reported in the published literature in an individual with breast cancer as well as in a reportedly healthy individual in a large scale breast cancer association study (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared/genes/BRCA2)). It was also described to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant.
