Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000539.3(RHO):c.165C>G (p.Asn55Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with lysine, which is basic and polar, at codon 55 of the RHO protein (p.Asn55Lys). This variant is not present in population databases (gnomAD no frequency). A different variant (c.165C>A) giving rise to the same protein effect has been determined to be pathogenic (PMID: 25359768). This suggests that this variant is also likely to be causative of disease. ClinVar contains an entry for this variant (Variation ID: 1353767). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt RHO protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic.
