NM_007294.4(BRCA1):c.3334G>C (p.Glu1112Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1112Q variant (also known as c.3334G>C), located in coding exon 9 of the BRCA1 gene, results from a G to C substitution at nucleotide position 3334. The glutamic acid at codon 1112 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
