Uncertain significance for Cranioectodermal dysplasia 2; Short-rib thoracic dysplasia 7 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020779.4(WDR35):c.1310C>T (p.Ala437Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with valine at codon 448 of the WDR35 protein (p.Ala448Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
