NM_005228.5(EGFR):c.2888A>G (p.Glu963Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E963G variant (also known as c.2888A>G), located in coding exon 24 of the EGFR gene, results from an A to G substitution at nucleotide position 2888. The glutamic acid at codon 963 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
